Amlodipine:
- A dihydropyridine calcium channel antagonist that inhibits calcium influx in vascular smooth muscle and cardiac muscle
- Acts predominantly on vascular smooth muscle, causing vasodilation and reduced peripheral resistance
- Decreases coronary vasoreactivity, lowers afterload, and reduces cardiac oxygen demand
Atenolol:
- A cardioselective beta-blocker (selectivity is dose-dependent)
- Lowers blood pressure by reducing cardiac output, suppressing plasma renin activity, and decreasing sympathetic outflow
- Provides cardioprotection by reducing myocardial oxygen demand through negative inotropic and chronotropic effects

- Usual dose: Amlodipine 5 mg + Atenolol 25 mg once daily
- If needed, dosage may be increased to two tablets daily or as directed by the physician

Disopyramide: Clearance reduced by 20% when combined; may enhance negative inotropic effects.
Ampicillin (≥1 g): May reduce Atenolol plasma levels.
Oral antidiabetics & insulin: Beta-blockers can reduce tissue sensitivity to insulin and blunt insulin secretion. Atenolol has relatively less impact, but caution is advised.

- Hypersensitivity to Amlodipine, Atenolol, or excipients
- Sinus bradycardia
- Second- or third-degree heart block
- Cardiogenic shock
- Hypotension
- Congestive heart failure or poor left ventricular function

Drug withdrawal: Avoid abrupt discontinuation; taper gradually under medical supervision to prevent rebound cardiac events.
